Dans le cadre de mon TP sur les bactériophages fécaux,
je dois observer la présence de batériophages
spécifiques d'E. coli sauvage dans l'eau d'un puits. La
première étape consiste en la culture d'E. coli qu'on
laisse incuber 3h, deux fois de suite avant d'enrichir l'erlen
contenant les bactériophages.
Est-ce qu'un quelqu'un
sait pourquoi la durée de l'incubation est de trois heures ???

Si je me réfère aux travaux de détection des
virus sur cellules eucaryotes, il est nécessaire de laisser la
culture démarrer (24 h) afin que toutes les cellules
adhèrent au support et les machineries cellulaires tournent à
plein tube. Dans le cas bactos, la génération est plus
courte (25mn) donc ce temps préliminaire de culture permet
d'infecter des cellules en pleine croissance et en bonne santé.
